Prediction of the Effect of Nucleotide Substitution Using Machine Learning

This thesis brings a new approach to the prediction of the effect of nucleotide polymorphism on human genome. The main goal is to create a new meta-classifier, which combines predictions of several already implemented software classifiers. The novelty of developed tool lies in using machine learning methods to find consensus over those tools, that would enhance accuracy and versatility of prediction. Final experiments show, that compared to the best integrated tool, the meta-classifier increases the area under ROC curve by 3,4 in average and normalized accuracy is improved by up to 7\,\%. Implementation of Sum Data Type in C++

Práce se zaobírá problematikou implementace součtového datového typu v jazyce C++. Nejprve navrhuje vlastní abstrakce pro manipulaci s posloupnostmi hodnot a typů, které následně umožňují vyjadřovat komplexní operace deklarativně. V průběhu implementace jsou prezentovány návrhové vzory obvyklé pro oblast metaprogramování. Implementace přichází s rozhodnutími odlišnými od referenční implementace. Pro manipulaci se součtovým typem jsou představeny dvě šablony funkce, lišící se sémantikou. Užití součtového typu je ukázano na příkladu konečného stavového automatu.
Design and Analysis of Reservation Teaching System

In this bachelor's thesis there is a detailed description of an information systems analysis and design. An attention paid to implementation of such system is also specified in further details. Techniques of procedure designing of these systems using UML are described on a specific case of reservation teaching system. This information system works with specific events, either a regular class or a one-time event. Particular users of this system are able to carry out actions, which are linked with event management as well as with subjects or classrooms.

The study of polymorphism and optimization of active pharmaceutical ingredients crystallisation

Active pharmaceutical ingredients (APIs) are frequently delivered to the patient in the solid-state as part of an approved dosage form (tablets, capsules, etc.). Understanding and controlling the solid-state chemistry of APIs is therefore an important aspect of the drug development process. APIs can exist in a variety of distinct solid forms, including polymorphs, solvates, hydrates, co-crystals and amorphous solids. Each form displays unique physicochemical properties that can profoundly influence the bioavailability, manufacturability, stability and other performance characteristics of the drug. Most APIs are purified and isolated by crystallisation from an appropriate solvent during the final step in synthetic process. The main objective of a crystallisation process is to produce crystals with desired properties such as particle size distribution (PSD), shape and purity. All pharmaceutical dosage forms must be produced in uniform units, and good content of uniformity is only possible when the size of the active component is carefully controlled. For on-line control of crystallisations of Quetiapine Fumarate to achieve desired PSD and no changed physicochemical purity was used the Lasentec Focus Beam Reflectance Measurement (FBRM) system.
Polymorphism of dicalcium silicate

The presented bachelor thesis is focused on the study of the mineral belite, polymorphism of belite, its conditions of origin and stability of polymorphs. The theoretical part is focused on the composition of cement, its minerals, modifications and properties. Furthermore, the possibilities of stabilization of polymorphs, synthesis and microstructure of dicalcium silicate are presented in the given section. The practical part is a summary of current research results at FAST BUT, THD. Based on previous work, a research was proposed in which a change in the properties of dicalcium silicate from a pre-fired mixture was observed.

GUI for REPOMO

Today, reconfigurable hardware is happend tendency when we can configure static circuit structure in the same way as software. REPOMO is a reconfigurable chip which contains 32 polymorphic gates. Polymorphism means that a gate can change its logic function as a response to the power supply voltage. The purpose of this thesis is to introduce the area of polymorphic circuits and construction and function of REPOMO chip. The main goal of the thesis is proposed and implemeted graphical user interface which make easy progress applications for chip REPOMO.
Utilization of lactate during exercise in the context of genetic dispositions

Title: Utilization of lactate during exercise in the context of genetic dispositions Objectives: The aim of the dissertation is to investigate, within a group of athletes from speed- endurance disciplines, the influence of selected polymorphisms in genes encoding lactate transporters MCT1-4 on lactate levels before, during, and after Intermittent 30s Wingate Test. A sub-objective of the study is to identify differences in allele and genotype frequencies in the respective polymorphisms between the group of athletes and controls. Methods: The research group consisted of 91 Eastern European Caucasian men, members of the Czech Athletic Association with elite performance in the 400 m run (age 20.4 ± 3.2 years; body weight 74.07 ± 7.5 kg; height 182.7 ± 6.14 cm). A control group (n=100; age 20.5 ± 1.2; body mass 77.5 ± 9.47 kg; height 181.5 ± 6.35 cm) of physically active individuals who did not perform speed and strength sports was used for allele and genotype comparisons. For genotypic analysis of selected polymorphisms, buccal mucosa swabs were performed in the probands. DNA isolation and subsequent genetic analyses were performed in collaboration with the Józef Piłsudski University of Physical Education in Warsaw. Variabilita genu MYOD1 ve vztahu ke kvalitě masa u prasat

This bachelor's thesis is dedicated to exploring the correlation between the MYOD1 gene and meat quality traits in pigs. The MYOD1 gene, which is a member of the MYOD gene family, is involved in muscle development during myogenesis. Therefore, mutations in this gene may affect meat quality. For the study 46 DNA samples of Czech Large White pigs were used, and genotypes were obtained using the PCR-RFLP method, which involved digestion of the PCR product using the DdeI restriction endonuclease. The frequencies of the genotypes were calculated, with the AA genotype representing 19,6 %, AB representing 47,8 %, and BB representing 32,6 %. The most frequent allele was B, which was represented in 56,5 % of the samples.Statistical analysis was performed using the SAS program. The results indicated that only the myoglobin content in 100 g of muscle was affected by the MYOD1 gene with a P ≤ 0,05 level of significance. Specifically, a statistically significant difference was observed between the AA and AB genotypes, with individuals carrying the AA genotype exhibiting significantly lower myoglobin content.
